Identification of Genes Expressed in Response to Dengue- 2 Virus infection of Culex pipiens quinquefasciatus (Diptera: Culicidae)

Abstract
To reveal expression of related genes of Culex pipiens quinquefasciatus (Say 1982): in response to dengue- 2 virus infection, differential display reverse-transcription PCR [DDRT-PCR] was used to identify molecular genetic changes in C pipiens quinquefasciatus after virus infection for 24 hs. Virus was isolated in C 6 / 36 cells from mosquitoes after 24 hs infection. Seven differentially expressed cDNA bands were identified in susceptible and refractory mosquitoes. Six of them were up-regulated and one was down-regulated following infection. Sequence comparison the GenBank showed that a high expressed gene fragment named as CD 16 A. The nucleotide similarity of CD 16 A with cDNA of ribosomal protein S 29 of Drosophila melanogaster and Mus musculus was 85 % and 79 %, respectively. The others were unknown gene fragments. Thus the fragment of CD 16 A should be the ribosomal protein S 29 gene of Cx pipiens quinquefasciatus. The GenBank accession number is AF 266749. The here presented findings may suggest that ribosomal protein S 29 gene was overexpressed in Cx pipiens quinquefasciatus indicating the restriction in host cell transcription. The result would be significant for understanding the mechanism of refractory in Cx pipiens quinquefasciatus to infection with dengue virus.
